To simplify ordering all RNAi oligo orders are synthesized with dTdT 3' ends and are annealed by moleculA unless otherwise specified.
To maximize your result, the following precautions should be taken when using siRNAs:
- If possible, knock-down your gene with two independent siRNA duplexes on varying regions of the mRNA target to control the specificity of the silencing process.
moleculA offers custom siRNA design for any target mRNA. Simply fill out the RNAi oligo order form* and paste your targets mRNA accession number or text sequence. Using the latest bioinformatics information available moleculA technical staff will design the best siRNA target site based on years of silencing expertise. Using powerful algorithms combined with known secondary structure features, moleculA is able to provide highly potent RNA oligos for RNA interference experiments. As with all our gene silencing designs, after receiving your order, all sequence information is provided to the user before we begin synthesis for verification.
Purification:
moleculA RNAi oligos are fully deprotected and purified via proprietary reverse phase mehtods. This method eliminates n-1 failure sequences, synthesis byproducts, and desalts the oligos. Reverse phase purification ensures that no cross contaimination occurs from other sequences. Purity is >90% and, in most cases, closer to 95%.
Scale:
Molecula currently provides RNAi oligo synthesis at the 50nmole, 200nmole, and 1.0 µmol scale. By default dTdT overhangs are added to the 3' ends. This typically provides a more reliable synthesis than UU as well as helps to protect the RNA from degradation. However you are free to choose either dTdT or UU overhangs.
