Designing antisense oligos that successfully bind to mRNA targets is the core service provided by Molecula. Using site-specific technology and years of experience, Molecula carefully identifies and selects sequences that suppress their intended targets.
In addition, sequences are screened for target specificity by BLASTING against known databases to assure they do not anneal to an unintended mRNA. Antisense sequences are also rigorously screened for motifs known to invoke immunostimulatory responses such as four contiguous G residues, palindromes of 6 or more bases and CG motifs.
Molecula modifies antisense oligos to prevent degradation by DNase present in cells or animals. Phosphorothioate, defined as the addition of sulfur to the oligo backbone, is the most common modification used to reduce degradation. This increased resistance results in greater levels of suppression.
Antisense oligos can provide a tremendous amount of fundamental data to committed investigators. Systematic down regulation of particular gene products can be key in determining a gene’s biological function and its role in a metabolic or regulatory pathway.

Oligonucleotides may be re-suspended in either sterile water or, preferably, TE (10mM Tris, pH 7.5-8.0, 1 mM EDTA). If you are making a concentrated stock solution we recommend dissolving your oligonucleotide in TE. Working solutions can be diluted from the stock solution using sterile, nuclease-free water (The use of DEPC-treated water is not recommended).
All Molecula Research Laboratories oligonucleotides are provided purified, lyophilized, and ready for use upon re-suspension.
Once re-suspended, oligonucleotide stock solutions are best kept frozen at -20 ºC for several weeks and may remain stable for several months. The most important factor in storing working solutions is using nuclease-free, sterile water. Drying down of your oligos and storing them at
-20 ºC is recommended for long-term storage.
Proper storage of your oligonucleotides will maximize their shelf life, allowing you to get the most use from your oligos.
Fluorescent oligonucleotides should be kept away from light as much as possible.
Alkaline phosphatase modified oligonucleotides should be stored at 4 ºC, never frozen.

Larger synthesis scales are available, please inquire. Our goal is to ship standard, desalted oligos within 24-48 hours. Oligos longer then 35 mer may take longer for delivery.
